Cytotoxicity of Crotalariae sessiliflorae L. against Human Oral Epithelioid Carcinoma Cells


Abstract
The cytotoxicity of Herba crotalariae sessiliflorae on cultured NIH 3T3 fibroblast and human oral epithelioid carcinoma cells were evaluated by 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) and Sulforhodamin B Protein (SRB) colorimetric assay method. The light microscopic study was carried out to observe- morphological changes of cultured mouse NIH 3T3 fibroblast (NIH 3T3) and human oral epithelioid cells (KB). The results were obtained as follows; In MT T assay, all extracts exhibited a significant cytotoxicity against human oral epithelioid carcinoma cells. Especially, hexane extracts showed low cytotoxic effect in NIH 3T3, but exhibited the strongest cytotoxicity in KB. The SRB absorbance in NIH 3T3 fibroblast was not significantly(P<0.05) decreased by treatment with ethyl acetate chloroform, and hexane extracts, but human oral epithelioid carcinoma cells was significantly(P<0.01) decreasd by treatment. The difference in SRB absorbance in two cell types was most remarkable when treated with hexane extracts. These results indicate that hexane extracts possess the cytotoxicity against human oral epithelioid carcinoma cells.
